The Five Killers erzählt die Geschichte von vier ehemaligen Auftragskillern, die sich am Grab ihres kürzlich ermordeten Anführers Knife (quasi Killer #5) treffen. Während die früheren Teamkollegen sich gegenseitig Vorwürfe machen und alte Wunden aufreißen, taucht ein komplett in weiß gekleideter Gast auf, der sich The Droid nennt. Droid behauptet Knife auf dem Gewissen zu haben und verspricht den übrigen Vier, dass sie ihrem ehemaligen Anführer bald ins Grab folgen werden. Um diesem Schicksal zu entgehen und den geheimnisvollen Droid zu erwischen, bevor er sie erwischt, müssen sich die Zerstrittenen noch einmal zusammenraufen.
