Knuckles ist ein roter, anthropomorpher Ameisenigel, der das Schatzsuchen über Alles liebt, wenn er mal nicht den Master Emerald auf Angel Island beschützen muss. Diese Aufgabe wird von dem Knuckles-Clan seit Generationen weitergeführt. Er ist der einzig Überlebende des Clans. Sonic und Knuckles sind Rivalen und zugleich auch gute Freunde, die sich gegenseitig unterstützen. Neben seiner Kraft und seinem Mut ist er aber auch naiv und dickköpfig, weshalb er auch leicht beeinflusst werden kann.
